Some brief advice on choosing the right capsid and promoter for your application:

Firstly, we are still learning about the extent of engineered AAVs for targeting diverse species. For many neuroscientists, working in C57BL/6 mice and targeting neurons, CAP.B10 and PHP.eB are often the best choice for an engineered AAV capsid. CAP.B10 displays lower liver targeting compared to PHP.eB and therefore increases the amount of virus that can cross through into the brain. However, other capsids show remarkable specificity for endothelial cells in brain, such as X1.1. AddGene has produced very helpful capsid selection guide that can be found at this link:

Getting the virus into the brain is only half the battle: the promoter/enhancer in use will have dramatic effects on the expression of your particular cargo. For example, the Allen Institute for Brain Science has done extensive work exploring different promoter/enhancers all within the context of PHP.eB. This characterization can be found here:
